What is a PCR plate?

In the polymerase chain reaction (Polymerase Chain Reaction, PCR), the PCR plate is mainly used as a carrier for the primers involved in the amplification reaction, Taq DNA polymerase, dNTP, template nucleic acid, Mg, buffer, etc. The PCR plate is available in transparent and milky white, among which milky white is more suitable for the new fluorescence quantitative PCR instrument that collects fluorescent signals at the top, but if the fluorescence quantitative PCR instrument is the bottom light source, it should be transparent. The PCR plate can be divided into three design modes: no skirt, half skirt, and full skirt according to the skirt design. The PCR plate is divided into 96 and 384 well plates into skirted and non-skirted plates. When using 96-well plates for real-time PCR experiments, it is recommended to use optical membranes to seal the reaction wells. How to seal it correctly? Press the membrane along the 96-well plate direction first, and then press along the transverse direction plate edge to seal it. After the sample is added, some small droplets will splash on the tube wall, and there will also be some air bubbles in the reaction system. Remove the air bubbles by centrifuging.

The PCR plate is a one-time consumable, and it is produced at one time. Cutting and then using it is not suggested, as it may cause experimental error and be partially reused or cannot be cleaned.

Precautions for the use of PCR plates

When using well plates for real-time PCR experiments, it is recommended to use optical membranes to seal the reaction wells. The correct sealing method is first to press the film along the longitudinal direction of the orifice plate, then laterally, and finally press along the edge of the plate to seal it. It should be noted that if the PCR tube cover is not properly covered, the PCR reaction solution will be thermally evaporated, which will affect the accurate control of the reaction temperature and the concentration of various components in the reaction, and will also become a source of pollution. Not only will some caps be deformed due to carelessness or uneven force during capping, but also cross-contamination of samples. Therefore, we recommend 8 tubes with independent caps. Each tube has an integrated cap to ensure the independent reaction of samples and avoid cross-contamination pollution.
